An investigation of the University of Murcia identify genetic changes that increase the risk of thrombosis in diabetics (21/01/2010)

Diabetes causes a deficiency of antithrombin (anticoagulant serpin) that contributes to the development of thrombosis among patients with chronic disease, according to a study by a faculty of the University of Murcia.

Microvascular thrombosis, both as macrovascular, is one of the major complications associated with diabetes and this research group has identified, in a paper published in "Thrombosis Research", one of the factors contributing to the hypercoagulability in diabetic patients.

Researchers have demonstrated the functional and conformational sensitivity of antithrombin, a serpin anticoagulant crucial in the hemostatic system, and have identified genetic changes that cause deficiency of this serpin and high risk of venous thrombosis.

The published work evaluated the functional and conformational nonenzymatic glycation associated with diabetes might have on antithrombin.

For this purpose, in vitro studies performed by incubating the purified protein or plasma with different concentrations of glucose and methyl glyoxal.

In diabetic patients, found that intracellular antithrombin during its folding and trafficking to its secretion into plasma was especially sensitive to this disease.

Under conditions of hyperglycemia, antithrombin is retained irregularly form aggregates, resulting in decreased secretion into the plasma and a slight deficiency.

This antithrombin deficiency intracellular retention in inclusion bodies which reduces the anticoagulant activity is an effect of diabetes, and could contribute to the development of thrombotic events observed in diabetic patients.

The research group belongs to the Department of Internal Medicine and, among its members, is Professor Vicente Vicente.
